Nature of Mutation
DNA sequencing using the SOLiD technique identified an A to G transition at position 1057 of the Chd2 transcript in exon 6 of 39 total exons. The mutated nucleotide causes a glutamine to arginine substitution at amino acid 162 of the encoded protein. The mutation has been confirmed by DNA sequencing using the Sanger method (Figure 1).
Protein Function & Prediction
The Chd2 gene encodes an 1827 amino acid proteinthat is a DNA-binding/helicase protein. CHD2 contains two Chromo domains at amino acids 260-346 and 376-449 (SMART).  These domains are found in proteins that alter chromatin structure. The protein also contains a DEXDc domain at amino acids 480-677, and other domains at amino acids 821-905 and 960-1244 typically found in helicases. Mice homozygous for a gene trap allele exhibit early postnatal lethality associated with fetal growth retardation. Mice heterozygous for a gene trap allele exhibit postnatal lethality and premature death after weaning associated with growth retardation and multi-organ defects.  
 
The Q162R change is predicted to be benign by the PolyPhen program.
