MGI Phenotype FUNCTION: [Summary is not available for the mouse gene. This summary is for the human ortholog.] The RAB5 protein is a small GTPase involved in membrane trafficking in the early endocytic pathway. The protein encoded by this gene binds the GTP-bound form of the RAB5 protein preferentially over the GDP-bound form, and functions as a guanine nucleotide exchange factor for RAB5. The encoded protein is found primarily as a tetramer in the cytoplasm and does not bind other members of the RAB family. Mutations in this gene cause macrocephaly alopecia cutis laxa and scoliosis (MACS) syndrome, an elastic tissue disorder, as well as the related connective tissue disorder, RIN2 syndrome. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jun 2011]

Nature of Mutation

DNA sequencing using the SOLiD technique identified a C to A transversion at position 392 of the Rin2transcript using Ensembl record ENSMUST00000110005 in exon 3 of 12 total exons. Multiple transcripts of the Rin2 gene are displayed on Ensembl and Vega. The mutated nucleotide causes a histidine to glutamine substitution at amino acid 52 of the encoded protein. The mutation has been confirmed by DNA sequencing using the Sanger method (Figure 1).

 
Protein Function and Prediction
The Rin2  gene encodes two isoforms of a guanine nucleotide exchange factor (GEF), and is necessary for activation of the RAB5B protein in the endocytic pathway. RIN2 also interacts with Ras, and forms homotetramers. Isoform 1 contains 903 amino acids, while the second isoform is missing the first 53 amino acids. An SH2 domain is located at residues 97-190, a VPS9 (vacuolar sorting protein 9) domain is located at amino acids 627-766, and a Ras-associating domain is found at residues 796-887. The protein also interacts with RAB5B at amino acids 506-775 (Uniprot Q9D684). No knockout phenotypes have been reported in mice. In humans, homozygous mutations in the RIN2 gene cause MACS syndrome (OMIM 613075), which is characterized by macrocephaly, alopecia, cutis laxa, and scoliosis.
 
The H52Q change is predicted to be possibly damaging by the PolyPhen program.
