Nature of Mutation
DNA sequencing using the SOLiD technique identified an A to T transversion at position 1605 of the Zeb1 transcript, in exon 6 of 8 total exons. The mutated nucleotide causes a histidine to leucine substitution at amino acid 529 of the encoded protein. The mutation has been confirmed by DNA sequencing using the Sanger method (Figure 1).
Protein Function and Prediction
Zeb1 encodes the 1117 amino acid zinc finger E box-binding homeobox 1 protein, also known as transcription factor 8 (TCF8). ZEB1 contains seven C2H2-type zinc fingers and one homeobox DNA-binding domain (Uniprot Q64318). ZEB1 is a zinc finger transcription repressor that binds to E-box sequences in the immunoglobin heavy chain enhancer, as well as the regulatory sequences of many tissue-specific genes. ZEB1 is known to repress T lymphocyte specific Il2 expression. In mice, mutations in Zeb1 affect thymus organization and homozygotes exhibit severe thymic T cell deficiency. Some mutations also exhibit extensive skeletal abnormalities. Homozygous mice generally die at birth. In humans, mutations in the ZEB1 gene result in posterior polymorphous corneal dystrophy-3 (PPCD3; OMIM #609141), a rare disorder involving metaplasia and overgrowth of corneal endothelial cells. ZEB1 inhibits expression of the collagen 4A3 (COL4A3) gene. Mutations in this gene cause Alport Syndrome (OMIM #203780), resulting in nephropathy and deafness.  
 
The D529G change is predicted to be probably damaging by the PolyPhen program. However, aoba homozygous mice are viable, and it is likely that this mutation is hypomorphic.
