MGI Phenotype FUNCTION: [Summary is not available for the mouse gene. This summary is for the human ortholog.] This gene encodes a central pair protein present in the axonemes of cells with a "9 + 2" organization of microtubules. The encoded protein is required for the proper function of the axoneme. Mutations in the orthologous gene in mice lead to primary ciliary dyskinesia characterized by immotile nasal and tracheal cilia, reduced clearance of nasal mucus, profound respiratory distress, hydrocephalus, and neonatal lethality within twelve hours of birth due to impaired airway mucociliary clearance. Single-nucleotide polymorphisms in this gene are associated with human height and targeted mutations lead to skeletal malformations affecting the limbs in mice, suggesting a role for this gene in skeletal development. PHENOTYPE: Homozygous null mice exhibit immotile respiratory cilia with axoneme structural defects, impaired mucociliary clearance, respiratory distress, pulmonary edema, disrupted alveolar epithelium, enlarged brain ventricles consistent with evolving hydrocephalus, failure to suckle, and neonatal lethality. Nature of Mutation

DNA sequencing using the SOLiD technique identified a T to C transition at position 96823 in the Genbank genomic region NC_000069 for the Spag17 gene on chromosome 3 (CTCGTTTCAG ->CTCATTTCAG). The mutation is located within intron 4, seven nucleotides upstream from the start of exon 5, and may impair the acceptor splice site of intron 4. Spag17 contains 48 exons. The mutation has been confirmed by DNA sequencing using the Sanger method (Figure 1).

Protein Function and Prediction
The Spag17 gene encodes the 2175 amino acid sperm-associated antigen 17 (SPAG17).   SPAG17 is important for the structural integrity of the central apparatus of the sperm axoneme, and contains two leucine-rich repeats (LRRs) at amino acids 1492-1521 and 1610-1633 (Uniprot Q5S003).
