MGI Phenotype FUNCTION: [Summary is not available for the mouse gene. This summary is for the human ortholog.] DEAD box proteins, characterized by the conserved motif Asp-Glu-Ala-Asp (DEAD), are putative RNA helicases. They are implicated in a number of cellular processes involving alteration of RNA secondary structure such as translation initiation, nuclear and mitochondrial splicing, and ribosome and spliceosome assembly. Based on their distribution patterns, some members of this family are believed to be involved in embryogenesis, spermatogenesis, and cellular growth and division. This gene encodes a DEAD box protein, which is a homolog of VASA proteins in Drosophila and several other species. The gene is specifically expressed in the germ cell lineage in both sexes and functions in germ cell development. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Oct 2009]
PHENOTYPE: Spermatogenesis is blocked in homozygous mutant mice, resulting in male infertility. Female mutant mice are fertile and do not exhibit any obvious reproductive defects. [provided by MGI curators]

Nature of Mutation

DNA sequencing using the SOLiD technique identified a T to G transversion at base pair 113404041 in the Genbank genomic region NC_000079 (Build 37.1) for the Ddx4 gene on Chromosome 13 (TTATCTATAG ->TTAGCTATAG). The mutation is located within intron 13 from the ATG exon, seven nucleotides to the next exon. The Ddx4 gene contains 22 total exons (Genbank record NM_001145885). Two transcripts of the Ddx4 gene are displayed on Ensembl.  The mutation has been confirmed by DNA sequencing using the Sanger method (Figure 1). 

 
Protein Function and Prediction

The Ddx4 gene encodes a 702 amino acid protein that plays a role in germ cell development. The protein is an ATP-dependent  RNA helicase and is expressed specifically in the testis. DDX4 contains an ATP-binding helicase domain at residues 292-475, and a C-terminal helicase domain at residues 503-648. A Q motif and a helicase DEAD box motif are present at amino acids 261-289 and 419-422, respectively. ATP-binding occurs at amino acids 305-312 (Uniprot Q61496). Isoform 1 (displayed on Genbank) contains 728 amino acids. Spermatogenesis is blocked in homozygous mutant mice, resulting in male infertility. Female mutant mice are fertile and do not exhibit any obvious reproductive defects.
