Nature of Mutation
DNA sequencing using the SOLiD technique identified an A to G transition at position 590 of the Pde4dip transcript in exon 5 of 46 total exons. Three transcripts of the Pde4dip gene are displayed on Ensembl. The mutated nucleotide causes a lysine to glutamic acid substitution at amino acid 257 of the encoded protein using Ensembl record ENSMUSP00000040905. The mutation has been confirmed by DNA sequencing using the Sanger method (Figure 1).
Protein Function and Prediction
The Pde4dip gene encodes a 2224 amino acid protein known as Myomegalin.  Myomegalin may function localize cAMP-dependent pathway components to the Golgi and/or centrosome, and contains a neuroblastoma breakpoint family (NBPF) domain of unknown function at amino acids 1497-1588 (Uniprot Q80YT7).
 
The K257E change is predicted to be benign by the PolyPhen program.
