FUNCTION: [Summary is not available for the mouse gene. This summary is for the human ortholog.] This gene encodes a member of the polycystin protein family. The encoded glycoprotein contains a large N-terminal extracellular region, multiple transmembrane domains and a cytoplasmic C-tail. It is an integral membrane protein that functions as a regulator of calcium permeable cation channels and intracellular calcium homoeostasis. It is also involved in cell-cell/matrix interactions and may modulate G-protein-coupled signal-transduction pathways. It plays a role in renal tubular development, and mutations in this gene cause autosomal dominant polycystic kidney disease type 1 (ADPKD1). ADPKD1 is characterized by the growth of fluid-filled cysts that replace normal renal tissue and result in end-stage renal failure. Splice variants encoding different isoforms have been noted for this gene. Also, six pseudogenes, closely linked in a known duplicated region on chromosome 16p, have been described. PHENOTYPE: Homozygous mutant embryos begin to die after embryonic day (E) 14.5. They develop edema by E13.5, pancreatic cysts by E15.5 and kidney cysts by E16.5. Heterozygous adults develop cysts of the kidneys (~20-30%) and the liver (~10%) late in life. [provided by MGI curators]

Protein Function and Prediction

Pkd1 encodes polycystin-1 (PC1), an integral membrane receptor that, in association with polycystin-2, functions to maintain normal renal tubular structure and function by acting as a mechano- or chemosensor in cilia and facilitating the PC2-mediated influx of calcium [reviewed in (1)]. PC1 has a large (i.e., >3000-aa) extracellular portion that contains sixteen immunoglobin-like PKD repeats, a receptor for egg-jelly (REJ) domain, and a G-protein-coupled receptor proteolytic site (GPS) that support protein-protein and protein-carbohydrate interactions [reviewed in (1)].  PC1 has 11 transmembrane domains and a 197-aa intracellular C-terminus [reviewed in (1)]. The intracytoplasmic tail is essential for signal transduction and mediates the ubiquitin-mediated degradation of PC1 [reviewed in (1)]. PC1 is proposed to act as an adhesion molecule that mediates cell-cell and cell-extracellular matrix interactions [reviewed in (1)].

Expression/Localization

PC1 is localized to the primary cilium, plasma membrane and the ER [reviewed in (1)]. In renal tubules, PC1 is at the basolateral membrane and in desmosomes and adherens juctions [reviewed in (1)]. PC1 expression is highest during kidney development but low in the adult kidney [reviewed in (1)].

Background

Mutations in PKD1 are linked to adult type I polycystic kidney disease [OMIM: # 173900; (2)], a condition marked by renal cysts, loss of glomerular filtration, end-stage renal disease, liver cysts, hypertension, and intracranial aneurysm.
