Nature of Mutation

DNA sequencing using the SOLiD technique identified a C to T transition at position 894 of the Tbc1d9 transcript in exon 2 of 21 exons using Genbank record NM_001111304.1.   Multiple transcripts of the Tbc1d9 gene are displayed on Ensembl and Vega. The mutated nucleotide causes a serine to arginine substitution at amino acid 57 of the encoded protein. The mutation has been confirmed by DNA sequencing using the Sanger method.

Protein Function and Prediction

The Tbc1d9 gene encodes a 1264 amino acid protein that may act as a GTPase-activating protein for Rab family members. The protein contains two GRAM domains at residues 146-213 and 293-361, a Rab-GAP TBC domain at residues 515-702, and a calcium-binding EF-hand domain at residues 886-921. GRAM domains are found in membrane-associated proteins (Uniprot Q3UYK3).
